THE INFLUENCE OF ASCORBATE ON ANTI-HUMAN MELANOMA ACTION OF A MONOLACUNARY WELLS-DAWSON NANOCLUSTER

ABSTRACT / SAŽETAK:

The influence of L(+)-ascorbic acid sodium salt (ascorbate), as a widely used antioxidant, on the anti-human melanoma A375 activity of a monolacunary Wells-Dawson polyoxometalate (mono- WD), α2-K10P2W17O61.20H2O was assessed in this study. A375 cells were in vitro exposed to the selected concentrations of mono-WD (the concentration range of 0.001 – 1 mM), ascorbate (0.040 – 40 mM), and the mixtures of mono-WD and ascorbate in the ratio of 1:40, for 24 and 48 hours. Both mono-WD and ascorbate induced concentration- dependent cytotoxic effects on A375 tumour cells, for both exposure times. The cell viability inhibiting potential of ascorbate was higher after 24-hour exposure, whereas similar cytotoxic effects of the studied compounds were obtained for 48- hour treatment. In summary, the results of the simultaneous exposure of A375 cells to mono-WD and ascorbate (mixed in the ratio of 1:40) showed that the presence of ascorbate does not affect remarkably the observed antitumour potency of the studied polyoxotungstate as a potential metallo- therapeutic.
