Fe(III)-CONTAINING WELLS-DAWSON POLYOXOTUNGSTATE AND ASCORBATE: EFFECT OF EQUIMOLAR MIXTURE ON HELA CELLS

САЖЕТАК / ABSTRACT:

This study aims to explore in vitro anti-tumor action of the equimolar mixture of Fe(III)-substituted monolacunary Wells-Dawson POM (Fe-WD), K7[FeIII2-P2W17O61)(H2O)] and L(+)-ascorbic acid sodium salt (ascorbate) using cervical carcinoma HeLa cells as a model system.

Exponentially growing HeLa cells were exposed in vitro to Fe-WD and ascorbate separately (0.005 – 0.5 mM) and the equimolar Fe-WD/ascorbate mixtures of the same concentrations for 48 and 72 h, at 37 °C. Single and simultaneous exposure to the studied compounds resulted in concentration- and time-dependent decrease of cell viability. IC50 values for Fe-WD were 0.015 and 0.0073 mM for 48- and 72-hour administration, respectively. Ascorbate induced noticeably weaker cytotoxicity with IC50 values of 0.37 and 0.32 mM for 48 and 72 h, respectively. Simultaneous exposure to the mixtures of Fe-WD and ascorbate demonstrated that the presence of ascorbate did not influence Fe-WD-induced cytotoxicity after the single treatment with the same concentrations for 48 and 72 h.
