САЖЕТАК / ABSTRACT:
The cytotoxic effect of the mixture of Fe(III)-substituted monolacunary Wells-Dawson polyoxometalate (Fe-WD), K7[FeIII(α2-P2W17O61)(H2O)] and L(+)-ascorbic acid sodium salt (ascorbate) on cervical carcinoma HeLa cells was evaluated in this study. Exponentially growing HeLa cells were treated in vitro with Fe-WD (0.001 – 1 mM), forty times higher concentrations of ascorbate (0.040 – 40 mM), and Fe-WD/ascorbate mixtures (concentration ratio 1 : 40) for 24 h, at 37 °C. Fe-WD and ascorbate inhibited cell viability in a concentration-dependent manner with different potencies. IC50 values were determined as 0.028 and 0.92 mM for Fe-WD and ascorbate, respectively. The simultaneous 24-hour exposure of HeLa cells to Fe-WD/ascorbate mixtures resulted in cytotoxic effects weaker than the mathematical sum of effects induced by single Fe-WD and ascorbate actions at the corresponding concentrations. Thus, it could be considered that Fe-WD and ascorbate act antagonistically on HeLa viability at the concentration ratio 1 : 40. This effect might result from the autoxidation of ascorbate in the presence of Fe(III) ions, resulting in ascorbate and Fe-WD consumption.
