SHOOT-TO-SHOOT VARIABILITY OF PLUM POX VIRUS DETECTION IN MICRPROPAGATED Prunus domestica L.

ABSTRACT / SAŽETAK:

The presence of Plum pox virus (PPV) was analyzed in the shoots of in vitro grown ‘Belošljiva’ plantlets using polymerase chain reaction (PCR) methods. Differences in detection between individual shoots and composite samples were evaluated in order to assess the reliability of virus diagnostics in in vitro material. Although virus titers in some shoots were below the detection limit, PPV was detected in at least one shoot in most plantlets, indicating stable transmission through micropropagation across successive passages. The small proportion of plantlets in which virus presence was not confirmed nevertheless calls for caution when designing experiments to evaluate antiviral treatments
